Abstract

The objective of the study is to formulate and evaluate the sustained release matrix tablets of glimipride using natural polymers tamarind seed mucilage and guar gum. Glimepiride is a first third generation sulphonyl urea agent for the treatment type II diabetes mellitus. The drug excipient mixtures were subjected to pre-formulation studies. The tablets were subjected to physicochemical studies, in-vitro drug release, kinetic studies and stability studies. FTIR and DSC studies indicated no chemical interaction between glimipride with the natural polymers used in this study. The physicochemical properties of tablets were found within the limits. The drug release from the optimized formulation was extended for a period of 12 h. As the concentration of polymers increased the drug release was inversely proportional. The optimized formulations were subjected to stability studies and shown there were no significant changes in hardness, friability and drug content. Results of the present study indicated the suitability of the using natural polymers tamarind seed mucilage and guar gum in the preparation of sustained release formulation of glimepiride.
